【文献解读】

β-淀粉样蛋白(Aβ)肽是阿尔茨海默病(AD)的标志,它会引起神经元一连串的氧化损伤,最终导致神经元死亡。2010年7月,江南大学的研究团队在期刊《Neurochemistry International》发表文章,探究了从中草药红景天中分离得到的红景天苷,对人神经母细胞瘤细胞(SH-SY5Y)抗Aβ诱导氧化应激的保护作用及其机制。

研究中诱导剂诱导的神经元毒性主要表现为细胞存活率下降、乳酸脱氢酶释放和神经元DNA凝聚。红景天苷预处理后,被诱导细胞的活力丧失和凋亡明显减轻。此外,红景天苷恢复了诱导剂诱导的线粒体膜电位的丧失,并抑制了细胞内活性氧水平的升高。红景天苷还可抑制诱导的c-Jun氨基末端激酶和p38 MAP激酶的磷酸化。

综上所述,红景天苷可减轻Aβ25-35诱导的SH-SY5Y细胞氧化应激和凋亡。红景天苷的保护作用不仅与调节内源性抗氧化酶和凋亡相关蛋白的表达有关,还与恢复异常的线粒体膜电位和细胞内活性氧水平有关。红景天苷对神经毒素的保护作用可能为其临床应用于治疗阿尔茨海默病的神经退行性变提供药理学依据。

Salidroside (p-hydroxyphenethyl-b-D-glucoside), which is extracted from Rhodiola rosea L. and has long been used as an adaptogen in traditional Chinese medicine, has been reported to have various pharmacological properties, including immunomodulation (Li et al., 2009), cardioprotection (Zhang et al., 2009), anticancer (Kucinskaite et al., 2004) and anti-oxidative effects (Kanupriya et al., 2005). In our previous study, we also reported that salidroside had protective effects against hydrogen peroxideinduced apoptosis in SH-SY5Y cells (Zhang et al., 2007). However, it remains undemonstrated whether salidroside exerts neuroprotection against Ab-induced neuronal damage. Considering the important role of Ab in the pathogenesis of AD, clarification of the effects of salidroside against Ab-induced toxicity may provide a new insight into its potential application to the prevention or treatment of AD.
